Product Introduction

Background

Accutase is a cellular digest containing protein hydrolase and collagenase activity. It is the perfect replacement for pancreatin/EDTA digestion fluid and is used to digest cells from conventional tissue culture vessels and adhesion culture vessels. Accutase is used to digest cells from conventional tissue culture vessels and adhesion culture vessels, and particularly suitable for tissue dissociation and cell separation. Because the separated cells can maintain intact surface antigens and do not contain any animal or bacterial source components, it can meet the requirements of subsequent experiments such as cell surface labeling, virus growth analysis, flow cytometry and bioreactor related tests.
Compared with the traditional pancreatic digestive solution, our product has the following advantages: ① Suitable for digestion of most primary cells and mammalian cell lines, as well as insect cells; ② Gentle and efficient digestion of stem cells (Human/Mouse Embryonic Stem Cells, hESCs and mESCs), higher cell viability after freezing; ③ The separation of adherent cells was achieved within a few minutes, no need for cleaning or neutralization reaction, saving cell passage time; ④ It has a mild effect on cell digestion, which can increase cell yield and survival rate; enhance cell adhesion efficiency; improve cell morphology and cell growth characteristics, etc.

Matters Need Attention

1. This product is only used for scientific research or further research, not for diagnosis and treatment.
2. This product was sterilized by 0.1 μm filtration and can be used directly after melting.
3. It is necessary to pay attention to the aseptic operation and avoid the contamination.
4. The product should be thawed at 2–8℃, do not melt this product at 37℃, otherwise it will affect the activity of the enzyme. Shaken thoroughly before use, when lesser amounts are required, aliquoting is advised to prevent repeated freeze-thaw cycles.
5. The digestion time can be adjusted according to the different reactions of different tissues or cells to Accutase solution. It is recommended to observe the digestion state under a microscope.
6. No additional washing and enzyme activity termination steps are required.

    Answer

    Accutase is a natural enzyme mixture derived from invertebrates (crustaceans), characterized by enzyme activities, including proteolytic and collagenolytic activity. Recombinant trypsin is a single serine protease produced through recombinant DNA technology in microbial expression systems; it mimics the function of conventional porcine or bovine trypsin while being animal-origin free. The core difference between Accutase and recombinant trypsin is that Accutase acts through the synergistic activity of multiple enzymes, whereas recombinant trypsin relies on a single enzyme activity.

  • Q:2 How long can trypsin solution be stored at 4°C?

    Answer

    We usually recommend that trypsin be placed in a 4℃ refrigerator and used within a week. If the amount used is small, trypsin can be divided into smaller packages as needed, and the unused portion can be stored in a -5~-20℃ refrigerator for long-term storage.

  • Q:3 How long can I keep trypsin in the refrigerator after thawing?

    Answer

    Generally speaking, trypsin can be stored refrigerated for up to a week. However, this may vary depending on how often it is warmed up and used during that week. It may also depend on how tightly the cells are attached, because there will be some loss of activity each day the trypsin is not frozen.

  • Q:4 What are the storage conditions for the Trypsin-EDTA (0.25%)?

    Answer

    We recommend storing the Trypsin-EDTA (0.25%) at  -5°C to -20°C as specified in the Storage section of our product page for this product. Although it is possible to store the trypsin for up to one week at +4°C, we would recommend preparing aliquots and storing them at -5°C to -20°C. These aliquots should be used up after thawing and freeze/thaw cycles should be avoided.
