Exosome Isolation Kit

Exosomes are a class of extracellular vesicles (EVs) with diameters of approximately 30-150 nm. They are secreted by various cell types under both physiological and pathological conditions and play a pivotal role in intercellular communication. Exosomes have demonstrated significant application potential across a wide range of research fields, including tumor microenvironment regulation, immune modulation, and neurodegenerative diseases.

Ultracentrifugation is the classical method for exosome isolation; however, its heavy reliance on specialized equipment and its time-consuming, labor-intensive procedures greatly limit research efficiency and accessibility. To address these challenges, the Procell R&D team has developed a series of Exosome Isolation Kits based on optimized separation strategies.   • Are the Exosome Isolation Kits suitable for both human and mouse samples?

    Answer

    Yes. The Procell Exosome Isolation Kits are not species-restricted and can be used with samples from humans, mice, and other mammalian species.

  • How can exosomes be isolated from 50 mL of bacterial culture supernatant obtained after high-speed centrifugation? Are there any recommended kits?

    Answer

    P-CA-502 or P-CA-503 can be used for exosome isolation.

    Due to the large sample volume, it is recommended to concentrate the supernatant prior to isolation. A 50 kDa ultrafiltration device may be used for sample concentration; please refer to the manufacturer’s instructions for detailed procedures.

    Alternatively, P-CA-505 can be used for simultaneous sample concentration and exosome isolation.

  • What target is used on the magnetic beads in the Micro Exosome Isolation Kit (Cat. No.:P-CA-501)?

    Answer

    The magnetic beads in this kit are functionalized to bind Phosphatidylserine (PS).

    PS is an important phospholipid that is normally located on the inner leaflet of the cell membrane. Exosomes, as nano-sized vesicles secreted by cells, are enriched in PS on their membrane surface, making PS an effective target for exosome isolation and enrichment.

    It should be noted that this method may also co-enrich membrane fragments derived from lysed cells.

  • Which exosome isolation kit is recommended for human umbilical cord mesenchymal stem cell-derived exosomes?

    Answer

    The choice of kit depends on the downstream application:

    P-CA-501 is recommended when extremely high exosome purity is required, such as for high-sensitivity molecular detection or omics analyses.

    P-CA-502/P-CA-503 are recommended for obtaining exosomes with high purity, intact structure, and well-preserved biological activity, suitable for cell function studies and drug delivery applications.

    P-CA-504 is recommended when experimental time is limited and the requirements for exosome purity and yield are relatively moderate.

    P-CA-505 is suitable for large sample volumes and cost-sensitive applications where downstream experiments do not require high purity. Please note that precipitation-based methods may co-precipitate impurities and potentially affect exosome properties.

    It is recommended to select the appropriate kit based on the sample type and downstream experimental objectives.
