Instructions 1. Check all containers for leakage or breakage.
2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation.
3. Suspension cells exhibit various morphologies, including single cells, irregular-shaped cells, and giant multinucleated cells.
4. A portion of cells will adhere to the culture vessel.
5. The cell line has numerous dark granules, and viability may range between 60% and 80%.

Subculturing Procedure Cultures can be maintained by either supplementing with fresh medium or exchanging medium via centrifugation. The recommended centrifugation conditions are 1200 rpm (approximately 250 × g) for 3 minutes.

Background CMK cells were established in 1985 from the peripheral blood of a 10-month-old boy with Down syndrome and relapsed acute megakaryoblastic leukemia (AML M7). They grow in suspension as large, pleomorphic single cells, with sporadic loose adhesion (<3%); 2–3% of the cell population are large, multinucleated cells. A moderate level of cellular debris accumulates during routine culture, and cell viability generally ranges from 60% to 80%.
