| Instructions | 1. Check all containers for leakage or breakage. 2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation. 3. Suspension cells exhibit various morphologies, including single cells, irregular-shaped cells, and giant multinucleated cells. 4. A portion of cells will adhere to the culture vessel. 5. The cell line has numerous dark granules, and viability may range between 60% and 80%. |

| Background | CMK cells were established in 1985 from the peripheral blood of a 10-month-old boy with Down syndrome and relapsed acute megakaryoblastic leukemia (AML M7). They grow in suspension as large, pleomorphic single cells, with sporadic loose adhesion (<3%); 2–3% of the cell population are large, multinucleated cells. A moderate level of cellular debris accumulates during routine culture, and cell viability generally ranges from 60% to 80%. |
