Handling information
| Instructions | 1. Check all containers for leakage or breakage. 2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation. 3. Some dark granules are present during cell culture. 4. Coated culture vessels are required for cell recovery; without coating, cells will float and fail to adhere after recovery. 5. Low-density seeding is recommended for cell recovery. 6. Cells exhibit predominantly adherent growth, with a trypsin dissociation time of approximately 4 minutes. 7. After subculture at a 1:2 split ratio, cells can be subcultured again around day 5–6. |

| Subculturing Procedure | 1. This cell line displays mixed adherent and suspended phenotypes. Although suspended cells remain viable, their density is low and they may be aspirated and discarded directly. 2. Rinse the adherent cells with PBS, then add 1–2 mL of 0.25% Trypsin-EDTA to the culture flask. Incubate at 37°C. Once the cells round up and detach from the vessel surface, add 4–6 mL of complete medium to terminate digestion. Gently pipette to generate a single-cell suspension, then harvest cells by centrifugation. 3. Seed the cells into fresh culture vessels at the recommended split ratio. |
