| Instructions | 1. Check all containers for leakage or breakage. 2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation. 3. Cells have a slow proliferation rate after recovery, and some floating material is present. At high cell density, the culture medium tends to turn yellow; perform timely medium exchange. 4. Dark granules, debris, and a dirty background are present; these can be removed by medium exchange. 5. When adherent cell density is high, it is recommended to focus on the adherent cell population; the suspended fraction can be removed by medium exchange. |

| Subculturing Procedure | 1. This cell line displays mixed adherent and suspended phenotypes. Although suspended cells remain viable, their density is low and they may be aspirated and discarded directly. 2. Rinse the adherent cells with PBS, then add 1–2 mL of 0.25% Trypsin-EDTA to the culture flask. Incubate at 37°C. Once the cells round up and detach from the vessel surface, add 4–6 mL of complete medium to terminate digestion. Gently pipette to generate a single-cell suspension, then harvest cells by centrifugation. 3. Seed the cells into fresh culture vessels at the recommended split ratio. |

| Background | HCC1569 is an epithelial cell line isolated from the breast tissue of a 70-year-old Black female patient with metaplastic breast carcinoma; the patient’s daughter also carries a germline mutation in the FHIT gene. Established in 1995, the cell line took 19 months to stabilize. The FHIT gene is located at chromosomal region 3p14.2, with a G-to-T transversion mutation at nucleotide 651. |
