Instructions 1. Check all containers for leakage or breakage.
2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation.

Subculturing Procedure 1. This cell line displays mixed adherent and suspended phenotypes. Suspended cells remain viable. Transfer the single-cell suspension into a centrifuge tube and pellet the cells via centrifugation at 1200 × g (approximately 250 × g).
2. Adherent cells can be dispersed by gentle pipetting to form a homogeneous single-cell suspension.
3. Mix all harvested suspended and adherent cell populations, then seed the cells into fresh culture vessels at the recommended split ratio.

Storage Conditions For long-term cryopreservation, cryovials should be stored in liquid nitrogen at −150°C to −196°C. Storage at −80°C is restricted to short-term interim use only.
Background HEL cells are a lymphoblastoid-like cell line derived from a 30-year-old Caucasian male with malignant erythroleukemia, capable of spontaneous and induced globin synthesis. HEL cells are negative for Epstein-Barr virus nuclear antigen (EBNA) and lack both surface and cytoplasmic immunoglobulins. HEL cells express HLA antigens (HLA-A3, Aw32, Bw35), β₂-microglobulin, and a proportion of the cells also express Ia antigens. HEL cells provide a model for studying erythroid differentiation and globin gene expression, analogous to murine Friend leukemia cells.
