| Background | The continuous cell line K-562 was established by Lozzio from the pleural effusion of a terminally ill 53-year-old female patient with chronic myelogenous leukemia. K-562 cells were originally classified as highly undifferentiated granulocytic cells; however, studies by Anderson et al. on their surface membrane properties identified K-562 as a human erythroleukemia cell line. K-562 cells are widely used as highly sensitive in vitro targets in natural killer (NK) cell assays. Pross et al. employed K-562 cells for the detailed in vitro detection of NK cells, including the mathematical quantification of NK cell activity. K-562 blasts are pluripotent hematopoietic malignant cells capable of spontaneous differentiation into recognizable erythroid progenitors, granulocytes, monocytes, and other lineages. K-562 cells are EBNA-negative. The K-562-GFP cell line was constructed from K-562 cells by lentiviral transduction and is a stable cell line capable of long-term, high-level GFP expression. This cell line exhibits low passage, high viability, and good condition, making it suitable for assays such as flow cytometry and fluorescence imaging. Selection drug concentration: Puro = 7.0 μg/mL. |
