| Instructions | 1. Check all containers for leakage or breakage. 2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation. 3. This cell line is a suspension cell line. Harvest the cell suspension by centrifugation; do not discard the culture medium directly. |
| Complete Medium | IMDM [PM150510]+10% Nutrient+1% Supplement |
| Incubation Atmosphere | Air, 95%; CO₂, 5% |
| Temperature | 37℃ |
| Subcultivation Ratio | 3×10⁵-5×10⁵ cells/mL |
| Medium Renewal | 2 to 3 times per week |
| Subculturing Procedure | Cultures can be maintained by either supplementing with fresh medium or exchanging medium via centrifugation. The recommended centrifugation conditions are 1200 rpm (approximately 250 × g) for 3 minutes. |
| Freeze Medium | General Freezing Medium [PB180436] |
| Storage Conditions | For long-term cryopreservation, cryovials should be stored in liquid nitrogen at −150°C to −196°C. Storage at −80°C is restricted to short-term interim use only. |
| Background | K-562 [K562] is a continuous cell line established by Lozzio from the pleural effusion of a terminally ill 53-year-old female patient with chronic myelogenous leukemia. Initially categorized as highly undifferentiated granulocytic cells, K-562 was later reclassified as a human erythroleukemia cell line following surface membrane property analyses conducted by Anderson et al. K-562 serves as a highly sensitive in vitro target widely utilized in natural killer (NK) cell functional assays. Pross et al. used K-562 to develop detailed in vitro detection methods for NK cells, including mathematical quantification of NK cell cytotoxic activity. K-562 blasts are pluripotent malignant hematopoietic cells that spontaneously differentiate into identifiable erythroid progenitors, granulocytes, monocytes, and other hematopoietic lineages. K-562 cells are EBNA-negative. |
| Tumorigenic | Yes, in nude mice (Tumors developed within 21 days at 100% frequency (5/5) in nude mice inoculated subcutaneously with 1×10⁷ cells). |
| Biosafety Level | BSL-1 |
