Instructions 1. Check all containers for leakage or breakage.
2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation.
3. This cell line is relatively difficult to recover. Some dead cells will be present after recovery, manifesting as dark granules in the background; the proliferation rate is slow.
4. An appropriate increase in IL-2 is recommended for weekend culture.
5. Minimize centrifugation during cell culture.

Subculturing Procedure Cultures can be maintained by either supplementing with fresh medium or exchanging medium via centrifugation. The recommended centrifugation conditions are 1200 rpm (approximately 250 × g) for 3 minutes.

Background KHYG-1 was established 1997 from the peripheral blood of a 45-year-old woman with aggressive natural killer cell (NK) leukemia; cells were described in the literature to display strong cytotoxic activity and to carry a point mutation in exon 7 of the TP53 (p53) gene.
