| Instructions | 1. Check all containers for leakage or breakage. 2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation. 3. The culture supernatant should be collected. |
| Complete Medium | RPMI-1640 [PM150110]+10% Nutrient+8 ng/mL Supplement1+1% Supplement2 |
| Incubation Atmosphere | Air, 95%; CO₂, 5% |
| Temperature | 37℃ |
| Subcultivation Ratio | 5×10⁵-1×10⁶ cells/mL |
| Medium Renewal | 2 to 3 times per week |
| Dissociation Duration | 1-2 min |
| Subculturing Procedure | 1. This cell line displays mixed adherent and suspended phenotypes. Suspended cells remain viable. Transfer the single-cell suspension into a centrifuge tube and pellet the cells via centrifugation at 1200 × g (approximately 250 × g). 2. Adherent cells can be dispersed by gentle pipetting to form a homogeneous single-cell suspension. 3. Mix all harvested suspended and adherent cell populations, then seed the cells into fresh culture vessels at the recommended split ratio. |
| Freeze Medium | 90% FBS+10% DMSO |
| Storage Conditions | For long-term cryopreservation, cryovials should be stored in liquid nitrogen at −150°C to −196°C. Storage at −80°C is restricted to short-term interim use only. |
| Background | M-07e is a cell line established in 1987 from the peripheral blood of a 6-month-old girl diagnosed with acute megakaryoblastic leukemia (AML M7) at initial diagnosis and represents a subclone of the growth factor-independent M-07 parental cell line. M-07e cells display proliferative responses to GM-CSF, IFN-α, IFN-β, IFN-γ, IL-2, IL-3, IL-4, IL-6, IL-15, NGF, SCF, TNF-α, and TPO. Notably, M-07e cells may rapidly acquire cytokine independence within 3–4 weeks of culture, likely resulting from the outgrowth of growth factor-autonomous subpopulations. Exome and RNA sequencing datasets are accessible for M-07e. |
