Instructions 1. Check all containers for leakage or breakage.
2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation.
3. This cell line is a suspension cell line. Harvest the cell suspension by centrifugation; do not discard the culture medium directly.

Background NCI-H209 [H209] cells were isolated and established in 1979 by A.F. Gazdar and colleagues from a bone marrow metastasis of a patient with small cell lung cancer. The bone marrow specimen was obtained prior to the patient's treatment. NCI-H209 cells are a classic small cell lung cancer cell line expressing high levels of four biochemical markers: neuron-specific enolase, brain-type creatine kinase, L-dopa decarboxylase, and bombesin-like immunoreactivity. The c-myc DNA sequence is not amplified, and no gross structural DNA abnormalities have been found. NCI-H209 cells synthesize p53 mRNA at levels comparable to those in normal lung tissue. NCI-H209 cells grow in suspension as aggregates; only cells within the aggregates are viable, and viability cannot be estimated. Typically, the culture medium contains a large amount of cellular debris.
