| Subculturing Procedure | 1. This cell line displays mixed adherent and suspended phenotypes. Suspended cells remain viable. Transfer the single-cell suspension into a centrifuge tube and pellet the cells via centrifugation at 1200 × g (approximately 250 × g). 2. Adherent cells can be dispersed by gentle pipetting to form a homogeneous single-cell suspension. 3. Mix all harvested suspended and adherent cell populations, then seed the cells into fresh culture vessels at the recommended split ratio. |

| Background | The NCI-H716 [H716] cell line was established from cells present in ascites fluid obtained from a patient following treatment with 5-fluorouracil. Unlike other colorectal cancer cell lines, NCI-H716 [H716] cells possess dopa decarboxylase and contain dense-core endocrine-type granules in the cytoplasm. NCI-H716 [H716] cells do not express TAG-72 or CA19–9 antigens, nor do they produce carcinoembryonic antigen (CEA). |
