Instructions 1. Check all containers for leakage or breakage.
2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation.

Subculturing Procedure 1. This cell line displays mixed adherent and suspended phenotypes. Suspended cells remain viable. Transfer the single-cell suspension into a centrifuge tube and pellet the cells via centrifugation at 1200 × g (approximately 250 × g).
2. Adherent cells can be dispersed by gentle pipetting to form a homogeneous single-cell suspension.
3. Mix all harvested suspended and adherent cell populations, then seed the cells into fresh culture vessels at the recommended split ratio.

Background The NCI-H716 [H716] cell line was established from cells present in ascites fluid obtained from a patient following treatment with 5-fluorouracil. Unlike other colorectal cancer cell lines, NCI-H716 [H716] cells possess dopa decarboxylase and contain dense-core endocrine-type granules in the cytoplasm. NCI-H716 [H716] cells do not express TAG-72 or CA19–9 antigens, nor do they produce carcinoembryonic antigen (CEA).
Tumorigenic Yes, in nude mice (Tumors developed within 21 days at 100% frequency (5/5) in nude mice inoculated subcutaneously with 1×10⁷ cells).
