Instructions 1. Check all containers for leakage or breakage.
2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation.
3. This is a suspension cell line. Both aggregates and single cells are present, and cells mainly grow in aggregates.
4. More single cells are present shortly after recovery.
5. Disperse cells as much as possible before cryopreservation.
6. Cells have a fast metabolic rate; timely medium exchange is recommended.
Complete Medium RPMI-1640 [PM150110]+10% Nutrient+1% Supplement
Incubation Atmosphere Air, 95%; CO₂, 5%
Temperature 37℃
Subcultivation Ratio 5×10⁵-1×10⁶ cells/mL
Medium Renewal Every 2 to 3 days
Subculturing Procedure Cultures can be maintained by either supplementing with fresh medium or exchanging medium via centrifugation. The recommended centrifugation conditions are 1200 rpm (approximately 250 × g) for 3 minutes.
Freeze Medium 90% FBS+10% DMSO
Storage Conditions For long-term cryopreservation, cryovials should be stored in liquid nitrogen at −150°C to −196°C. Storage at −80°C is restricted to short-term interim use only.
Background The RC-K8 cell line was established in 1984 from the ascites of a patient with lymphoma. The disease was originally classified as true histiocytic lymphoma, at an advanced, refractory stage. This cell line was later reclassified as the GCB-like lymphoma subtype (germinal center B-cell type). The cells harbor a BCL6 translocation.
Biosafety Level BSL-1
