Instructions 1. Check all containers for leakage or breakage.
2. Remove the frozen cells from the dry ice packaging and immediately transfer them to liquid nitrogen (liquid or vapor phase) for long-term cryopreservation.

Subculturing Procedure 1. This cell line displays mixed adherent and suspended phenotypes. Suspended cells remain viable. Transfer the single-cell suspension into a centrifuge tube and pellet the cells via centrifugation at 1200 × g (approximately 250 × g).
2. Adherent cells can be dispersed by gentle pipetting to form a homogeneous single-cell suspension.
3. Mix all harvested suspended and adherent cell populations, then seed the cells into fresh culture vessels at the recommended split ratio.

Storage Conditions For long-term cryopreservation, cryovials should be stored in liquid nitrogen at −150°C to −196°C. Storage at −80°C is restricted to short-term interim use only.
Background SHP-77 retains important characteristics of small cell lung cancer (SCLC) and possesses stable biochemical properties, allowing continuous cultivation. This cell line is an unusual undifferentiated large-cell variant of SCLC. Although it exhibits a variant morphology, it maintains the biochemical properties of classical SCLC. Electron microscopy reveals gland formation and the presence of intracytoplasmic lamellar bodies. These cells express the neuroendocrine marker L-dopa decarboxylase and contain dense-core secretory granules. SHP-77 can serve as a target for detecting cytotoxicity via ⁵¹Cr and ¹¹¹In release in both in vitro assays and in vivo nude mouse models, and is used to evaluate the immune reactivity of cells and serum from lung cancer patients. This cell line can also be used to assess the immune status of SCLC patients undergoing radiotherapy or chemotherapy.
Tumorigenic Yes; Yes, the cells form tumors in athymic nude mice, and usually grow as circumscribed nodules without evidence of metastases.
