| Subculturing Procedure | 1. This cell line displays mixed adherent and suspended phenotypes. Suspended cells remain viable. Transfer the single-cell suspension into a centrifuge tube and pellet the cells via centrifugation at 1200 × g (approximately 250 × g). 2. Adherent cells can be dispersed by gentle pipetting to form a homogeneous single-cell suspension. 3. Mix all harvested suspended and adherent cell populations, then seed the cells into fresh culture vessels at the recommended split ratio. |

| Background | The TF-1 cell line was established in 1987 by T. Kitamura et al. from a heparinized bone marrow aspirate obtained from a 35-year-old Japanese male with severe pancytopenia. TF-1 cells are strictly dependent on IL-3 or GM-CSF for long-term growth and do not respond to IL-5. Various lymphokines and cytokines, including IL-1, IL-4, IL-6, IL-9, IL-11, IL-13, G-CSF/GM-CSF, LIF, and NGF, exert biological effects on TF-1 cells. The cells do not express glycophorin A or carbonic anhydrase I. Their morphological and cytochemical characteristics, together with constitutive globin gene expression, indicate that TF-1 cells are committed to the erythroid lineage. |
