Product Introduction

Background

M199 (Medium 199) originally used in nutritional studies of chicken embryo fibroblasts, it has been widely used in the culture of various animal cells, including some non-mammalian cells. M199 is especially suitable for the culture of untransformed cells, rat pancreatic epithelial cells and mouse lens tissues, also used in virology and vaccine production. Compared with other basic medium, M199 contains unique ingredients, including adenine, adenosine, hypoxanthine, thymine, and other vitamins. There are two kinds of equilibrium salt (earle's salt and hank's salt) in M199. Earle's salt is often used in CO₂ environment, hank's salt is used in non-CO₂ environment.
This product contains many kinds of amino acids, vitamins, inorganic salts and other ingredients for cell culture, but does not contain protein, lipids or any growth factors. Therefore, the product should be used with serum or serum-free additives.

FAQs

  • Q:1 What temperature should be used when preparing liquid medium from dry powder medium?

    Answer

During the preparation of liquid medium from dry powder medium, dissolution is typically carried out at room temperature (approximately 20–30 °C). This temperature range is sufficient to ensure proper dissolution without compromising the stability of the medium components.

  • Q:2 Are there any specific requirements for the sodium bicarbonate used when preparing powdered culture media?

    Answer

Analytical-grade sodium bicarbonate is sufficient. Please note that after preparation, the powdered culture medium should be filter-sterilized.

  • Q:3 Why do we need to add sodium bicarbonate to dry powder culture medium? Can’t we add it to the dry powder in advance and dissolve it directly during preparation?

    Answer

Sodium bicarbonate (NaHCO3) in cell culture medium is usually not included in the dry powder form of culture medium, but is added as a separate component during the preparation of culture medium solution. The main reasons include: 1. Sodium bicarbonate is easy to decompose under certain conditions and is sensitive to carbon dioxide. If it is pre-mixed into dry powder culture medium, it may affect its stability and cause the effective ingredients to fail. 2. Since the carbon dioxide concentration in different laboratories and the specific conditions (such as temperature and time) of the operator during preparation may be different, sodium bicarbonate is added during the final preparation of liquid culture medium. Its concentration can be adjusted according to actual needs to ensure that the culture medium reaches the ideal pH value. 3. Storing sodium bicarbonate separately can also facilitate the transportation and long-term storage of dry powder culture medium, because sodium bicarbonate may cause caking when it is damp, affecting its solubility.

  • Q:4 How much temperature control is needed during the preparation of liquid media from dry powdered media?

    Answer

Dissolving can be controlled at room temperature (20-30°C).
